rSD is blocked by the 5-HT(1D) receptor agonist sumatriptan and by the NMDA receptor antagonist MK-801. Drugs were injected i.v. 10 min before initiation of photothrombosis at concentrations of 3 mg/kg sumatriptan, 3 mg/kg MK-801, and 10 mg/kg memantine. (a) Sumatriptan and MK-801 decreased the incidence of wave generation during the first hour after photothrombosis. Memantine, another NMDA receptor antagonist, trended towards reducing the incidence of wave generation. (b) MK-801 decreased the size rSD waves. (c) MK-801 reduced rSD wave propagation velocity. (d) None of the drugs affected the site of rSD wave initiation. Regardless of treatment group, waves were predominantly initiated at the optic disc. For all panels, n = 5 animals per treatment group.
