Quality of preexisting F3-specific CD4 T cells impacts the Ad5-F3 boosting effect. C57BL/6 mice were primed as indicated to generate different qualities of F3-specific CD4 T cells and 6 weeks later boosted by injection of Ad5-F3. Sixteen weeks after the boost, spleens were removed and single-cell suspensions prepared. Cells were incubated with the F3 protein and analyzed by flow cytometry for expression of CD154 or the indicated cytokine (A). Data are means and SEM (n = 5 per group).
