ChpArec phosphotransfer to Xpts under conditions of ChpArec saturation.
A, single time point analysis for reactions containing BeF3− as a sensitive detection of ChpArec to Xpt phosphotransfer. The reactions contained 6 μm ChpA −HD, 40 μm Xpt, 70 μm BeF3− (70 μm BeCl2 and 10 mm NaF), and 30 μm [γ-32P]ATP and were incubated for 90 min before quenching and analysis. B, reactions contained 7.3 μm ChpA −HD, 56 μm HPt, and 30 μm [γ-32P]ATP. After 10 min, 2 mm unlabeled ATP was added; samples were removed at designated times for quenching and analysis.
