Treatment with a proteosomal inhibitor increased expression of DNMT1, and TGF-β1 treatment decreased DNMT1 ubiquitination. CCL210 cells were treated with TGF-β1 (2 ng/ml) for 4 or 8 h in the presence of the proteosomal inhibitor MG-132 (10 μm). Cell lysates were immunoprecipitated for DNMT1 or DNMT3a, resolved by SDS-PAGE, and probed with antibodies against ubiquitin and DNMT1. A, representative blot of ubiquitinylated DNMT1 is shown.
