Mean traces for the time-dependent change in [Ca²⁺]_i in Fura-2-loaded Trpm4^+/+ and Trpm4^–/– PCMCs are presented as the ratio of fluorescence at 340 nm to that at 380 nm (340/380). n = 3 independent experiments with at least 50 cells per single measurement were pooled for the analysis. Cells were incubated over night with 300 ng/ml anti-DNP-IgE and stimulated with 100 ng/ml DNP at the indicated time period.
