rWNT5A partially rescues the EMT changes induced in HB2 cells. a Immunofluorescence staining for E-cadherin and vimentin was performed for the WNT5A rescue experiments. In these experiments, HB2 cells that were transiently transfected with WNT5A siRNA were treated with rWNT5A (0.4 μg/ml). For visualization, a secondary goat anti-mouse/-rabbit Alexa-488-labeled antibody was used together with DAPI counterstaining. The magnification used was 60X (n = 3). b Representative Western blot demonstrating a reversal in the expression of the epithelial markers E-cadherin and vimentin in rWNT5A-treated cells compared with cells treated with WNT5A siRNA alone. c An invasion analysis was performed using a Transwell assay, in which WNT5A siRNA-treated cells were plated on Transwell invasion inserts in either the presence or absence of rWNT5A. Vehicle-exposed cells were used as negative controls. Statistical comparisons were made with Student’s t-test. All error bars represent the standard error of the mean (n = 4). *p < 0.05
