The 20S pre-rRNA accumulates in the ubi3ΔN mutant. Strains TLY56.D3 (UBI3-HA) and TLY63.A3 (ubi3ΔN-HA) were grown in YPD medium at 30 or 22°C to an OD₆₀₀ of 0.8. Total RNA was extracted from each strain. Equal amounts of RNA (5 μg) were subjected to northern hybridization. (A) Northern blot analysis of high-molecular-mass pre- and mature rRNAs. (B) Northern blot analysis of low-molecular-mass pre- and mature rRNAs. Signal intensities were measured by phosphorimager scanning; values (indicated below each panel) were normalized to those obtained for the wild-type control, arbitrarily set at 1.0.
