(A) Acidic granules (arrowhead) were tracked by incubating the cells with 100 nM red LysoTracker® DND-99 (LT) for 24 h at 37 °C. After fixation, cells were observed under bright field (BF), UV illumination for oocyst autofluorescence and DAPI staining of the macrophage nucleus, and the appropriate filter set for LT fluorescence. Arrow indicates LT-oocyst colocalization. Macrophages cultured in absence of oocysts served as negative controls. Scale bar: 10 μm. (B) The number of LT-positive macrophages were determined over 800 cells counted per experiment. ***p < 0.001 (n = 4).
