VP22 is not sufficient to inhibit antigen presentation by CD1d following an HSV-1 infection. (A) VP22/GFP- or control GFP-expressing HEK293-CD1d cells were lysed and resolved by SDS-PAGE. The expression of VP22 was detected by western blot analysis using a VP22-specific IgY antibody. The arrow points to the expected VP22 band. (B) VP22/GFP-expressing HEK293-CD1d (solid line) or GFP-expressing HEK293-CD1d cells (dotted line) were stained for intracellular CD1d HC and the surface CD1d-β2m complex as indicated, and analyzed by flow cytometry. (C) VP22-expressing HEK293-CD1d and control cells were cocultured with human NKT cells for 48 h. The supernatants were harvested for a GM-CSF ELISA.
