Viral protein synthesis is important for VP22-mediated inhibition of CD1d-mediated antigen presentation. HEK293-CD1d (VP22/GFP) and control (GFP) cells were mock infected or infected with ΔUL49 for 6 h at an MOI of 5, with or without cycloheximide. The cells were subsequently fixed and cocultured with human NKT cells for 48 h. Data are presented as the percentage of GM-CSF produced by NKT cells relative to mock treatment (set at 100%).
