Cytoplasmic 8-OHdG immunoreactivities were co-localized with mitochondrial protein VDAC immunoreactivities. Upper panels: sham cardiomyocytes. Middle panels: failing cardiomyocytes. Bottom panels: failing cardiomyocytes exposed to ISO (10 nM) and the mitochondria-targeted ROS scavenger Mito-tempo (100 μM). B. Bar graph representation of the data in Fig 2A. Relative mitochondrial DNA damage = (fluorescence intensity of 8-OHdG) / (fluorescence intensity of VDAC). The bars indicate the means ± SE. Each group included 20–30 cells. At least 6 cells were evaluated for each preparation.
