Relaxed pBR322 DNA was mixed and incubated with GyrA, GyrB, ATP and FQs at the indicated concentrations. Each FQ was examined for its inhibitory activity against WT DNA gyrase and mutant DNA gyrases with Gly89Cys-GyrA (indicated as Gly89Cys), Ala91Val-GyrA (Ala91Val) and Asp95Gly-GyrA substitution (Asp95Gly). The lanes labeled as R indicate relaxed pBR322 DNA.
