(A) Two independent CWR22Rv1 p54/nrb knockout (KO) clones #11 and #24 were used for rescue experiments. Re-expression of p54/nrb significantly increases the PCGEM1 level. (B) Re-expression of p54/nrb significantly increases the AR3 level, as determined by Western blot. (C) Ectopic expression of PCGEM1 in p54/nrb KO #11 cells increases AR3 as detected by qRT-PCR. The exogenous p54/nrb was tagged with Myc. (D) Re-expression of p54/nrb enhances tumor cell growth. KO #11 and #24 cells were transduced with pCDH-Myc vector (vector) or pCDH-Myc-p54/nrb (p54/nrb) and then plated out in 96-well plates in the absence of androgen.
