Cabazitaxel induces non-apoptotic cell death in A549 cells. (A and B) A549 cells were exposed to 5 µg/ml cabazitaxel or 20 µM 5-fluorouracil (5-FU) for 24 h at 37°C, then stained with Annexin V/PI or terminal deoxynucleotidyl transferase dUTP nick end labeling-fluorescein isothiocyanate (FITC)/propidium iodide and 4′,6-diamidino-2-phenylindole (DAPI), and were then subjected to flow cytometry or fluorescence imaging. Magnification, ×10. (C) Cabazitaxel (5 µg/ml) or 5-FU (20 µM) treated A549 cells were harvested and lysed, and the collected supernatants were subjected to caspase 3 activity assay. (D) A549 cells were pretreated with 20 µM z-VAD-FMK for 1 h, were incubated with cabazitaxel (5 µg/ml) or 5-FU for 24 h, and cell death was subsequently determined by trypan blue dye exclusion assay. Normal non-treated A549 cells were used as the control. Data are presented as the mean ± standard deviation of three independent experiments *P<0.05, **P<0.01.
