Inflammatory monocytes are TLR9-responsive cells and accumulate in peripheral tissues following CpG-induced inflammation. Yet40 (IL-12 reporter) mice were treated with or without 50 μg of CpG1826 for four doses and leukocytes were isolated from tissues. (A) Cells were stimulated with CpG and the induction of YFP was measured by flow cytometry. YFP+ cells were further characterized by their expression of Ly6C and Ccr2 and plots show representative data of three separate experiments. Percentage (B) and total numbers (C) of Ly6GLy6C+Ccr2+ inflammatory monocytes were enumerated in mice treated with and without repeated TLR9 activation. Graphs show data from seven to eight mice per group and they were analyzed by Mann–Whitney U tests. Numbers of significance symbols correspond with the P value (i.e., 2 symbols, P < 0.01; and 3 symbols, P < 0.001).
