CDDO-Me and 268 do not suppress ErbB2 transgene expression. A, E18-14C-27 cells, derived from a MMTV-ErbB2 tumor (7), were treated with compounds for 48 h or MMTV-neu mice with established tumors were fed control diet, CDDO-Me (60 mg/kg diet), 268 (100 mg/kg diet), or the combination for 1 wk, and lysates were analyzed by Western blotting. B, E18 cells were incubated with CDDO-Me (0.3 μmol/L), 268 (1 μmol/L), or the combination for 72 h, and cell proliferation was measured with a [3H]thymidine incorporation assay. *, P < 0.05 versus control. C, to examine apoptosis, E18 cells were treated for 48 h, and Annexin V and propidium iodide staining were measured by flow cytometry.
