Both FAK inhibition and Latrunculin-A prevented actin polymerization during capacitation. (A) To determine the effects of FAK inhibition and Lat-A on actin polymerization, guinea pig spermatozoa were capacitated in the presence or absence of PF573228 (5 µM) or Lat-A (50 µM). F-actin was stained using TRITC-labeled phalloidin. Images are representative of three independent experiments. (B) Fluorescence levels of TRITC-labeled phalloidin were quantified using the software Nis 3.1 to define the time course of F-actin during capacitation in the presence or absence of PF573224 or Lat-A (mean±s.e.m., n=5 independent experiments).
