HepG2 cells were cultured for 6 h with 50μM Amitriptyline (Amit) in the presence of CoQ, Vitamin E, BHA or NAC. A. ROS production was analyzed at 6 h by Mitosox staining and flow cytometry as described in the Materials and methods section. B. Mitochondrial membrane potential was analyzed at 6 h by flow cytometry using MitoTracker Red. Data represent the mean±SD of three separate experiments. *P < 0.01, between control and Amitriptyline treated cells; ap < 0.01, between the absence or presence of antioxidant.
