HA-Ca_V1.2_e22 or Ca_V1.2_e21+22 were co-transfected with α₂δ and β_2a subunit in HEK293 cells with or without MG132 treatment. β_2a subunit was cloned in pIRES2-EGFP (as an indicator of β_2a subunit expression). Immunostaining of total Ca_V1.2 channels and confocal imaging were performed after 48 h transfection. (A) β_2a subunit-expressing cells showed up-regulation of total HA-Ca_V1.2_e22 channels with or without MG132 treatment, compared to cells not expressing β_2a subunits. (B) Total Ca_V1.2_e21+22 channels were not markedly altered in β_2a subunit-expressing cells with or without MG132 treatment. Scale bar, 20 μm.
