(A,B) Immunofluorescence images showing the location of the (A) FM4-64 (1 uM; cell membrane marker) and subcellular p47 phox, or (B) subcellular p47 phox and TRPM2, in fixed cells by using confocal microscopy. The cells were pre-treated with 3,4-dihydro-5-[4-(1-piperidinyl)butoxy]-1(2H)-isoquinolinone (DPQ; 100 μM) under low glucose (LG; 5.5 mM glucose;) or high glucose (HG; 30 mM glucose). The percentage of co-localization of p47 phox with (A) FM4-64, or (B) TRPM2, was calculated as the average volume of the overlapping areas (n = 4–5).
