Pedigree structures include wide families (A), deep families (B), and small families (C) (Figure S1). In this simulation, 40% of rare variants are protective with an OR of 0.25, whereas 60% are deleterious with an OR of 4. We assume that individuals in the top two generations are not genotyped. Power was measured at α = 0.05 from 2,000 replications of simulations.
