TPD52 is a target gene of miR-139-5p in uterine leiomyoma cells.

Notes: (A) Bioinformatic study showed that 3′UTR of TPD52 contains a conserved binding site for miR-139-5p. (B) miR-139-5p overexpression inhibited the expression of TPD52 mRNA. Expression of mRNA was determined by RT-PCR (***P<0.001). (C) miR-139-5p overexpression inhibited the expression of TPD52 protein. miR-139-5p mimics or NC treated cells were incubated for 72 hours. Total protein was extracted and TPD52 expression was determined by Western blot. (D and E) Dual-luciferase reporter assay validated the TPD52 as a target of miR-139-5p (**P<0.01).

Abbreviations: NC, negative control; RT-PCR, real-time polymerase chain reaction; 3′UTR, 3′ untranslated region; WT, wild type; Mut, mutant type.
