Immune responses to i.a. injection of p5RHH-p65 NP. (A) Mice were injected i.a. with the indicated NP or HBBS (no Tx); their serum was obtained on day 14 and probed for the presence of IgG or IgM antibodies against p5RHH-p65 siRNA nanocomplexes. The levels of specific antibody response were measured by direct absorbance at 450 nm. (B) Mice were injected i.a. with HBSS or p5RHH-p65 siRNA NP, and their liver was collected at 6 h; total RNA was prepared and probed for the expression of IFN gene response 54 (ISG54) by real-time PCR. Poly(I:C) injected i.v. served as positive control for TLR stimulation and type I IFN response. Data represent mean ± SEM; n = 3–5 mice per treatment group.
