Cell attachment assay of HCVcc- and patient-derived HCV samples. (A) Cell attachment of sh-NT medium, sh-apoE medium, and apoE3HA medium-mixed HCV. Huh7.5.1 cells and viruses were precooled to 4°C. Attachment was performed on ice for 0.5 h and 2 h. After attachment, cells were washed 5 times with cold PBS, followed by TRIzol treatment and RNA extraction for qRT-PCR. (B) Huh7.5.1 cells and patient-derived HCV (samples 1, 2, and 3) were precooled to 4°C. Attachment was performed on ice for 2 h. After attachment, RNA quantification was conducted as described above. (C) HCV was mixed with sh-NT medium, sh-apoE medium, and apoE3HA medium, followed by incubation at 37°C for 4 h. The resulting samples were used to perform a heparin column attachment assay. After 1 h of attachment, the heparin column was washed 4 times with cold PBS and subjected to TRIzol treatment and RNA extraction for qRT-PCR.
