PBMCs from control donors (C) and at-risk individuals (ARIs) (14–17 per group) were stimulated with anti-CD3 for 24 hours. The expression of indicated cytokines was quantified with ELISA or real-time PCR (A). The levels of cytokines from ARIs who are ACPA+ and ARIs who are ACPA are compared in B. In addition, the transcript levels of PFKFB3, ATM, and G6PD measured with real-time PCR are shown in C, and the concentration of lactate in supernatant is shown in D. The transcript levels of PFKFB3 and ATM in ACPA+ and ACPA ARIs are compared in E.
