(a) Cell lysates from human tumour cell lines growing anchorage-dependently were immunoblotted for the indicated proteins. (b) Tumour cell lines were grown anchorage dependently (2D), as multi-cellular tumour spheroids (Sph) for 10 days or in mimetix 3D scaffold (Mim) for 14 days. Cell lysates were immunoblotted with the indicated antibodies. (c) Tumour cell lines growing anchorage-dependently were treated with 1 μM Chk1i for 24 hours. Cell lysates were immunoblotted for the indicated proteins. Expression levels of pChk1 (S296), total Chk1, Cdc6 and CDT1 were determined by densitometry and from Figs 1b and 2a.
