Representative traces of sensor responses to cellular zinc perturbation with TPEN (100 µM) and pyrithione (5 µM)/ZnCl2 (10 µM) for sensor mutants 2 (a), 3 (b), 4 (c), 5 (d) and 6 (e) with different linker regions. Sensor 2 and 5 showed unstable maximal signal with treatment of zinc, while 4 demonstrated the most stable maximal intensities (indicated by the red circle).
