Effects of hypoxanthine on cholesterol levels and lipogenesis‐related genes in APOE‐deficient HepG2 cells. (A) HepG2 cells were transfected with APOE‐siRNA (siAPOE) or non‐target‐siRNA (siN) for 24 hrs and then treated with hypoxanthine (1 mM) for 24 hrs. Total cholesterol and cholesteryl ester levels were assayed in cell lysates. (B) APOE,ABCA1,HMGCR and LDLR mRNA levels were determined by real‐time RT‐PCR. (C) Representative western blot for APOE, ABCA1, HMGCR, LDLR and β‐actin is shown. (D) Relative protein expression was determined using densitometry.
