(A) Full MS scan prior to isolation and fragmentation of intact H4 containing an acetyl and dimethyl group. The isolation window is set so that the species of interest is the most abundant ion in the scan so that it will be selected for data-dependent MS2 acquisition. (B) Fragment ion coverage of the H4 protein containing N-terminal acetylation (AC), K16ac (AC), and K20me2 (DI). There are fragment ions flanking each modification, allowing for confident identification of the PTMs. (C) Deconvoluted MS/MS spectra.
