HA consists of alternating disaccharide units (D-glucuronic acid and N-acetyl-glucosamine) connected by glycosidic bonds. The sHA1 derivative (D.S. 1.0–1.5) is preferentially sulfated at C6 of the N-acetyl glucosamine unit. The labeling of HA and sHA1 was performed with ATTO565-NH₂ and resulted in a side-on modification of carboxylic groups (A). The double-labeled ATTO565-ATTO655-sHA1 derivative was generated by end-on modification with ATTO565-NH₂ followed by side-on functionalization with ATTO655-NH₂ (B).
