Trx activity was measured by an insulin disulfide reduction assay and defined as the increase in absorbance at 650 nm per min. V_max and K_m values were determined by non-linear regression using the Prism 5 program. tTrx1, wild-type selenoprotein T. denticola Trx1; tTrx1/U32C, Sec-to-Cys mutant of tTrx1; tTrx1/U32W, Sec-to-Trp mutant of tTrx1; tTrx2, Cys-containing T. denticola Trx2; eTrx1, E. coli Trx1. ND, activity not detectable.
