A. 293T cells were transiently transfected with HA-BMI1 (left panel) or pRS-shBMI1 (right panel). After 48 hrs, the total cell lysates were prepared from transfected cells and the expression of BMI1, p16 (a BMI1 target) and β-actin was detected by Western blot analysis using specific antibody against each protein. B. Downregulation of miR-141 and miR-200c by BMI1 overexpression (left panel), and upregulation of miR-141 and miR-200c by BMI1 knockdown (right panel) were confirmed by qRT-PCR in the above set of cells as described in the Methods.
