The 3′utr sequence of Psen1 (A), Psen2 (D), and Notch1 (G), predicted to include relative miRNA MREs, were aligned and both wt and mt sequences are listed. Wt and mt plasmids were cotransfected with relative miRNAs for 24 h, and luciferase reporter activities were analyzed for miR-375 toward Psen1 (B), miR-30a toward Psen2 (E), and miR-34a toward Notch1 (H). Protein levels were determined following overexpression or knockdown of miR-375 (C), miR-30a (F), or by miR-34a (I). Data shown are means ± SEM and representative of three separate experiments.
