Figure 6. — Schematic illustration of Set7 dependent transcriptional network in mESC differentiation. In its repressed and undifferentiated state, the Set7 promoter is characterized with a bivalent domain and enriched with OCT4 and SOX2. Following ESC differentiation OCT4 and SOX2 are released from the Set7 promoter for transcriptional activation associated with H3K4me3. Increased Set7 protein promotes the broad expression of differentiation-associated genes (SMC, Heart, Adipose, Muscle and Brain). The results of experimental studies performed here indicate Set7-dependent control of SM-associated genes by SRF methylation and H3K4me1.
