Correlation of thermosensing activity in Escherichia coli with thermodynamic stability. (A) Western blot analysis of CssA-GFP expression. Total protein isolated from indicated strains were subject to western blot analysis. Membranes were probed with antibodies recognizing GFP or RecA. Nucleotide shown in black box is removed in the respective sequence whereas the nucleotide in red box is mutated to U. Their respective melting temperature is shown (in°C) at the bottom of secondary structure. The blue colored residues in CssA6 are mutated/deleted in CssA71, CssA72 and CssA75. (B) Thermosensing properties of the CssA RNA constructs averaged over three biological replicates. The bar chart shows average expression of GFP relative to the loading control RecA, together with their standard deviations.
