An experimental approach for the transcriptional analysis of distant enhancer-promoter communication in chromatin. A. Restriction map of the pYP05 template. Positions of the enhancer, promoter and 13 high-affinity nucleosome positioning 601 sequences (601 NPS) are indicated. 35 bp of the promoter-proximal NPS is truncated from its promoter-distal end and is replaced by bacteriophage T7 transcription terminator site, limiting the length of the transcript to 176 nucleotides.
