(A) Representative isothermal titration calorimetry (ITC) thermograms showing that all three Cascade/I-C subunits exhibit low micromolar affinity for each other in the absence of crRNA. Affinities are reported as KD ± standard error of the fit parameter (SE). Three independent experiments for each interaction yielded the following values for Cas5c-Cas7: KD = 3.8 ± 1.3 μM, n = 0.59 ± 0.082, ΔH = 5,100 ± 2,200 cal/mol, ΔS = 8.6 ± 7.5 cal/mol/deg; Cas5c-Cas7: KD = 3.9 ± 0.80 μM, n = 0.57 ± 0.14, ΔH = 4,800 ± 520 cal/mol, ΔS = 9.4 ± 1.7 cal/mol/deg; and Cas7-Cas8c: KD = 6.0 ± 1.9 μM, n = 1.0 ± 0.30, ΔH = 5,200 ± 610 cal/mol, ΔS = 7.4 ± 2.3 cal/mol/deg.
