TGF‐β1 partially abrogates the effect of FLCN knockdown on chemotaxis and contraction of 3‐D collagen gels in HFL‐1 cells. HFL‐1 cells were transfected by a lentivirus that expressed shRNA against FLCN (shFLCN1) or control shRNA (control). FLCN‐knockdown reproducibly impaired both chemotaxis and gel contraction by HFL‐1 (P < 0.01). After transduction, HFL‐1 cells were maintained in either complete media without (−) or with TGF‐β1 (+) at 10 pmol/L and then subjected to assays for chemotaxis (A) and 3‐D collagen gel contraction (B) assays.
