Effect of SOD1 mutation on cell growth and viability. (A,B) Wild-type (WT) or cells deleted for SOD1 containing either an empty plasmid (control) or re-expressing SOD1 or sod1 mutants as indicated were assessed for growth using an automatic plate reader. (C) Viability was assessed after 24 h of growth in selective medium and assessed using a colony forming unit assay, which calculates the percentage of cells able to form a colony within each population. The probability of a significant difference in viability compared with wild type was calculated for each strain; ***P<0.001. All experiments were carried out in biological triplicate and error bars represent standard deviation.
