Rg1 protects against H2O2-induced neuronal apoptosis through myosin IIA. (A) PC12 cells were pre-incubated with blebbistatin (1 μM), Y27632 (10 μM) or z-VAD-fmk (10 μM) in the presence or absence of 10 μM of Rg1 for 12 h, and then treated or untreated with 100 μM of H2O2 for another 12 h. Caspase-3 activity was assayed by caspase-3 activity assay kit. Results were expressed as mean ± SD from three independent experiments (##P < 0.01 versus vehicle-treated cells, *P < 0.05 versus H2O2 treated cells, **P < 0.01 versus H2O2 treated cells). (B) Proposed binding site of Rg1 in myosin IIA. This work used four docking programs based on different matching and shape matching algorithms to assure the accuracy of the final scores and the precision of conformers.
