Adjustment of hybridization stringency for miR-34a in fresh brain tissue. ISH of miR-34a was performed in fresh samples treated with 1 μg/mL proteinase K. (A) miR-34a probes were hybridized at the indicated temperatures (55°, 48°, and 37°C) followed by washing with 50% formamide in 1 × SSC at the hybridization temperature. (B) The sections hybridized at 48°C were washed with buffer solution including the indicated percentages of formamide (70, 50, and 30%) in 1 × SSC.
