(A) Human sperm were preincubated in capacitation medium for 3 h. 8-Br-cAMP (1 mM) was added for the last 10 min of the preincubation, and PMA (25 nM) was added for another hour. Sperm lysates were analyzed for ERK2 activity by western blotting as above. A representative blot is shown and bars are mean ± SEM from three experiments. (B) Human sperm were preincubated in capacitation medium for 3 h. 8-Br-cAMP (1 mM) was added for the last 10 min of the preincubation, and PMA (25 nM) was added for another hour. The percentage of acrosome-reacted cells was determined using FITC-conjugated Pisum sativum agglutinin (PSA-FITC) as described in methods. The data represent the mean ± SEM of duplicates from 3 experiments. Means designated by * and ** are significantly different (p < 0.05 and p < 0.01), respectively.
