Workflow of cisplatin-seq. Following DNA fragmentation, cisplatin-modified DNA fragments are enriched by domain A of HMGB1. Cisplatin–DNA adducts cause primer extension (first strand DNA synthesis) to stop at the sites of cisplatin crosslinking, which gives base-resolution information of cisplatin crosslinking sites.
