a) Janus kinase (JAK)2/signal transducer and activator of transcription (STAT)1 phosphorylation was stimulated by beryllium in peripheral blood mononuclear cells (PBMCs) from chronic beryllium disease (CBD) patients. PBMCs were treated with 100 μM BeSO4 for the indicated period of time and cell lysates containing equal amounts of total proteins were analysed by Western blot using antibodies against phospho (p)-JAK2, total JAK2, p-STAT1, STAT1 and actin. Total JAK2 and STAT1 served as a loading control. b) The JAK2 inhibitor TG101348 downregulated lymphocyte proliferation in CBD (n=6). PBMCs were incubated in the absence (“Medium”) or presence of 1 μM TG101348 for 30 min, and treated with without or with 10 and 100 μM BeSO4 for 4 days. Data are presented as mean±SEM stimulation index (SI).
