Schematic of the first proofreading step. The ribosome exits the initial selection step with rate constant kc (Fig. 1) and forms the complex, RT3GDP, in which the ribosome is bound to aa-tRNA in ternary complex with EF-TuGDP. The ternary complex aa-tRNAEF-TuGDP may dissociate from the ribosome with rate constant q1c in cognate and q1nc in near-cognate cases. Alternatively, T3GDP splits with rate constant kTu into a ribosome-bound aa-tRNAEF-TuGDP complex in which aa-tRNA and EF-TuGDP do not interact and where the two molecules are held in place by their respective affinities to the ribosome. Complex aa-tRNAEF-TuGDP may reform ribosome-bound complex aa-tRNAEF-TuGDP with rate constant qF1 or form ribosome complex Raa-tRNA through dissociation of EF-TuGDP from the ribosome with rate constant kF1.
