Figure 6

Altered dynamics of DSBs in xnd-1 are mys-1-dependent. Dissected germ lines were stained with DAPI to visualize DNA and anti-RAD-51 to visualize DSB repair intermediates. The beginning of the transition zone is marked with a solid line, the end with an arrowhead; onset of significant RAD-51 foci accumulation with a dashed line. (A) Kinetics of DSBs are altered in xnd-1 compared to wild-type, with high levels of RAD-51 appearing in the transition zone. DSB accumulation in mys-1 is similar to wild-type, peaking in midpachytene. mys-1 suppresses the formation of early DSBs in xnd-1. (B) Ectopic expression of him-5 cannot suppress the induction of early DSBs in xnd-1. RAD-51 accumulation appears normal in eaIs15 animals (top). xnd-1 is epistatic to eaIs15 and the double mutant shows accumulation of RAD-51 foci in the transition zone. Scale bar, 20 μm. DAPI, 4’,6-diamidino-2-phenylindole; DSB, double-strand break.
