Interaction with p300 modifies SRY DNA-binding activity. The GST/SRY (wt) and (K136R) fusion proteins were bacterially expressed and purified. Equivalent amounts of SRY proteins were incubated in in vitro acetylation reaction in the presence (+) or absence (−) of p300 and acetyl CoA and submitted to binding reaction in EMSA experiments with ³²P-labelled TCF probe. Reaction products were resolved by electrophoresis and visualised by autoradiography. Specific SRY complexes are indicated.
