By hydrolyzing genomic DNA to bases, deoxycytidine and deoxy-5-methylcytidine would become Cyt and 5mCyt, the molecular weights of which were 111 and 125 respectively. Thus, they could be discriminated by mass spectrometer, indicating the methylation degree of genomic DNA. 100 MCF7 cells in water were injected in the tip of a nanopipette before being hydrolyzed.
