LRP1 is a key receptor for heparanase reuptake into ECs. RAOEC and RHMEC lysates were used to determine the expression of LRP1, n = 7 and n = 4 (A and B). In RAOEC, siRNA was used to silence LRP1, followed by determination of myc-Hep^L uptake and conversion to Hep^A, n = 5 (C–E). RAOEC were pre-treated with or without 200 nM RAP (F) or 20 μg/mL LRP1 neutralizing antibody (G) for 1 h prior to incubation with 500 ng/mL myc-Hep^L for 4 h. Cell lysates were collected to determine Hep^L uptake, n = 5. 20 μg/mL IgG was used as a control for the LRP1 neutralizing antibody experiment. *P < 0.05, **P < 0.01, ***P < 0.001.
