Spt6 and FACT are required to suppress tgp1+ expression. (A) Histone density over the tgp1+ locus was measured by histone H3 ChIP-qPCR experiments in phosphate-replete wild-type cells, Stp6 mutant cells (spt6-1), and cells lacking the activity of the FACT subunit Spt16 (spt16-18) grown for 1 h at the restrictive temperature of 36°C. (B) Northern analysis of tgp1+ mRNA levels in wild-type cells, 1343Δ cells with constitutive tgp1+ expression (i.e. cells lacking the promoter that drives repressive nc-tgp1 transcription), and in cells with compromised Spt6 or FACT activity grown at the restrictive temperature for 1 h. (C) RT-qPCR analysis of tgp1+ mRNA levels in phosphate-replete wild-type cells, spt6-1 cells, and spt16-18 cells grown at the restrictive temperature for 1 h.
